描述: | Alzheimer's disease is a common neurodegenerative, progressive, and fatal disorder. Generation and deposition of amyloid beta (Aβ) peptides associate with its pathogenesis and small soluble Aβ oligomers show the most pronounced neurotoxic effects and correlate with disease initiation and progression. Beta amyloid 1-42 is a 42-amino acid peptide which plays a key role in the pathogenesis of Alzheimer, even slightly increased amounts of this Aβ(1–42) might be sufficient to cause Alzheimer's disease (AD). | ||||

1. 将Aβ肽(1-42)粉末溶于冷的六氟-2-丙醇(HFIP)。肽溶液在室温下孵化至少1小时,以建立单体化和随机化的结构。(Solid Aβ peptide was dissolved in cold hexafluoro-2-propanol (HFIP). The peptide was incubated at room temperature for at least 1h to establish monomerization and randomization of structure.) 2. 蒸发掉HFIP,将得到的多肽以薄膜形式保存在-20或-80℃。(The HFIP was removed by evaporation, and the resulting peptide was stored as a film at -20 or -80 ℃.) 3. 将得到的薄膜以5 mM的浓度溶解于无水DMSO中,然后用涡旋方式稀释到适当的浓度和缓冲液(不含血清和酚红的培养基)中。(The resulting film was dissolved in anhydrous DMSO at 5 mM and then diluted into the appropriate concentration and buffer (serum- and phenol red-free culture medium) with vortexing.) 4. 接下来,该溶液在4-8℃下老化48h。然后在4-8 ℃下以14000g离心10分钟;可溶性低聚物在上清液中。将上清液稀释10-200倍用于后续实验。(Next, the solution was age 48h at 4-8 ℃.
